Does it matter how long you mix?

No effects at all. You can just double-click it for Mixed drinks.

Slow (short time) shaking = mixing; quick (long time) shaking = blending

I seem to recall that in the prologue the longer you mix a drink (assuming you're not meant to blend it) without blending it the better the quality of the drink is but I've seen nothing to that effect here. It probably doesn't matter and if it does it probably affect how much tips you get I guess.

I'm actually beginning the game and it's not explained, I had to come here to understand that there was a different between the two type of movements.

You are mixing until the shaker speeds up, at which point you are blending. I've found shake the shaker 10 times (so, back and forth 5 full times) is perfect for mixing. I'm not sure what happens if you mix for too long. I always stop maybe a second or two after it the shaker speeds up, to make a blended drink.
