Not a lot to go off. What mods are they? How did you install them (manual/workshop/mod manager)? Could you have accidentally tried to install classic edition mods to definitive edition? Or tried installing to the wrong edition?

Check the '..\Documents\Larian Studios\Divinity Original Sin 2 Definitive Edition\Mods' folder to see if the pak file is there. A few people have reported issues in the Definitive Edition of the game with Steam not copying mods to the Documents folder.
If that is the case, you can search the '..\SteamApps\workshop\content\435150' folder for *.pak files and copy the recent ones there manually.

If that doesn't help, try deleting the modsettings.lsx file in your profile folder. The game should then recreate it the next time it is started, from the mods in the My Documents folder.
Deleting that file is also useful if you've added a bunch of mods in multiple sessions, and want to get them all in alphabetical order without manually reordering them (and you don't care about having to re-enable any mods you are using in that profile).
