Only the host can load it up to play. The host can do that, and play alone controlling every character in the party if they want. Or they can load it up, and invite the other person to join, and then the host can use the menu option to assign control of the party members to the players. They can assign them the same way they played before or give control of different party members to different people if they want.
The game does not treat the save file any differently. You can play it single player or coop. The game don't care.
You can never take a character out of your game and jump into some other game with it. And if you are not the host, the only way you can play it is if the host invites you, or if they email you a copy of the save. But then it becomes two different games, one for each of you.
