"highest quuality" is subjective, so we don't know what you're talking about without an explanation.

If you're focused on resolution, then use a search engine to find what resolutions and aspect ratios work best for Instagram. For example, a 1:1 square image might be 1080 pixels wide by 1080 high.

Then do the math to scale down to an appropriate pixel art resolution, say 108 x 108, which is 1/10th scale. Save your art to an .aseprite file while you work on it.

When you're finished, export it to a file format accepted by Instagram with nearest neighbor upscale. In the example, 10x would be the inverse of 1/10x. Try to avoid file formats that use lossy compression, like jpg or jpeg; for some social media sites, this may not be possible.

If you're focused on wide color gamut, HDR color, etc., then use different software. Aseprite works best with standard RGB (sRGB), fixed range, 8 bits (1 byte) per color channel.
