Click on the button above the palette swatches with an icon of an arrow pointing down. A menu will appear. Select Gradient from the menu. The colors in-between the poles will update.
If you are using Aseprite v1.3 beta, there is a Gradient by hue option in the arrow menu.
This feature is not as good as it could be. You can see that for yourself if you make Gradient by hue from a gray to cyan or if you make a Gradient from green to red. If you're not satisfied with the colors generated by Aseprite, search online for a gradient or palette generator that uses a perceptual color space.
Other Aseprite palette management features are in the menu to the right of the down arrow menu, see the button with the three horizontal lines icon.
