You will have another chance. Citizens are not glowing anymore only if the eavesdrop sequence has been seen.
But there's at least one circumstance where you've only one chance because it's triggered by a quest. There might be some more like that.
During the quest regarding the cure for Harvey Fiddick in the hospital you can give the placebo cure to Dr. Ackroyd. Afterwards he's going upstairs and you can eavesdrop on him. At least until now i never saw him go upstairs again since then - and i was actually waiting in the hospital for a pretty long time
