Unless Jack locks you in a room for a 1-1 fight, don't waste ammo on him. Headshot if you must to knock him back, and run in the opposite direction. (Quickturn helps.) He's just like Mr. X/Nemesis from the remakes.
The "black gooey thingies" aka The Molded are down for the count. However, they respawn later, based on your progression in the game. Thumb rule: if the enemies don't go down after a good shotgun shell to the head, just run the other way. You may already know this, but reload from checkpoint, if you wasted too much ammo.

Not sure if you need to restart. The game will shower you with bullets. This game has less things to shoot than any other RE game... and yet you will find an unreasonable amount of ammo throughout. Even if you wasted bullets on him, you probably can still make it.
But then again.. you are still at the beginning... so if you feel like restarting.. why not? It's not a long game anyway.
The molded can be killed for good and you should kill them. They don't exactly respawn.. but new ones may appear where there were none in different places as you play through the game.

Other than that I had well over 200 Machine gun bullets, a few hundreds flamethrower ammo and a couple of flame grenade. Well over 10 chem fluids and 3 or 4 enhanced chem fluid. A few bottles of first aid and super first aid and well over 10 herbs.
That's on Madhouse and I killed pretty much every enemy up to that point. Except some few enemies not worth bothering with.
