In the True Lab... [spoilers]
Is there anything significant about that giant red goat-looking skull? On the sign beside it, it says "DT Extraction Machine" which I guess stands for Determination Extraction Machine. I mean, I guess it would be obvious Alphys used it to get determination, but it kind of just freaks me out, particularly the way it looks.

The only thing I've seen about it (besides the diaries telling you what it is) is that God/Omega/whatever people call him Flowey's head is actually the same shape as the DT machine while he's trying to cyriak you to death.

it's the shell that Flowey fights you in, in the nuetral endings.

Goat skulls are a reoccurring symbol in Undertale. I want to associate it with Toriel/Asgore/Asriel, because they're goatlike monsters, but... in the No Mercy ending when you fight sans, he has these goat-skull shaped lasers. The sprite/attack name comes up as "Gaster Blaster".
There are a lot of theories surrounding Gaster, but it's all pretty much speculation. Datamining and tinkering with the "Fun" settings in the source files can trigger rare NPCs that talk about him.
They say that Gaster used to be the old Royal Scientist disappeared when he fell into his own invention and was "shattered across space and time".
However, the goat-skull shaped DT machine doesn't seem like something Gaster was studying, nor does it seem like he would have been smeared across space-time via the determination extractor... so perhaps it was an old machine of his that Alphys repurposed.

Of course, this is all speculation.

Most of the discussion regarding the DT machine is speculation, since nothing is really explained in the game.

There are two people that seem to have similar symbols, Omega Flowey and Sans. Sans makes sense that he would use a DT extraction tactic, considering during the Genocide run, his entire job is to kill you. Omega Flowey... I have no idea, since he shouldn't have any real ties to it at all.
