Technically.
If you beat the game without killing anything you can just reload your save and do the true pacifist content. Otherwise you have to start a brand new playthrough and not kill anyone that time. Either way you gotta beat the game first.
Unless you're actually trying to get one of the thirteen varations of the neutral ending, it's a good idea to just go for true pacifist on your first try. Saves you the pain of having to start over.
At any rate, never do Genocide first. I'm not saying you can't do Genocide, I'm just saying save it for last. There's a TON of reasons I could list as to why you should save it for last.
