Why do Sans & Asgore both know this?
Sans & Asgore can both tell how many times someone has died. In a genocide run, Sans can tell how many times he's killed you by the look on your face. In other runs, he says you haven't died a single time but, if you have, he says something like "What's that look? Am I wrong?"

If you tell Asgore he's killed you before, he nods sadly & how sadly he nods depends on how many times he's killed you. Maybe Chara died more than once & that's how Asgore knows but how does Sans know? What kind of♥♥♥♥♥♥has Sans seen? Or done?

Regardless, this makes Asgore's game over dialogue about Chara staying determined & waking up because they're the future of humans & monsters seem more significant.

That is a good question... I honestly think it has something to be with strongest monsters or something else.

They don't just know it immediately. Your answer is already in what you typed. Sans examines your expression to know if you've died before and you just simply tell Asgore how many times he's killed you before. The game over screen is only playing along with the game's design to trick new players into thinking they were naming the protagonist at the beginning.

If I told you you killed me 5 times, you wouldn't nod miserably. Nodding is an admission of knowledge.
