Pacifist first because it will teach you the mechanics of the game whereas genocide it expects you to be advanced in the mechanics of the game. Plus pacifist has a much better story and Genocide won't be understood without having completed a full pacifist play through.

Pacifist is best for after Neutral. While it is hard at some points, it has a great storyline and you won't feel terrible and have a bad time like in Genocide.

Pacifist is much better than the genocide route. That´s because you see much more in the story of the underground and the characters. And the battles against the monsters are harder than in the genocide route (after a few minutes of play you kill 90% of the monsters with one hit). A other reason you should choose the pacifist route is, that killing all monsters gets boring because you are constantly busy making an encounter with walking around. The most people I now are making the genocide for one reason: The fight with Sans (you can´t call this really a spoiler, because the internet is full with him!). And after you completed the genocide route, you have to fiddle with the memory data until you want to play the pacifist route, without a nasty surprise at the end.

There are only two challenges: Undyne and Sans. They are hard bosses, but the other bosses (and normal monsters are die with one hit). Maybe the game is a bit more difficult at the two spots than in the pacifist run, but otherwise it is much easier.

You're missing the entire point of the game. You're supposed to play it your own way. Kill the characters you don't like and spare the ones you like.
