Have you tried restarting Steam, then running Steam as Admin (right-click on Steam), and then Verifying the Integrity of the Game Cache Files by right-clicking on Smite in Steam and going to Properties and the Files tab?
You could also try right clicking on Smite in Steam (again, while running as Admin as above), then Properties and the Files tab again, but this time click Browse Local Files, then try Binaries > Redist > InstallHirezService.exe. It will probably ask you to uninstall and reinstall HiRez service again, which will take longer than the verification, but this should hopefully fix it if the verification step didn't.
That might not work, but that's where I usually start for any issues I run into with Smite.
You can also use this guide to reinstall through Steam, following all the same instructions, just installing through Steam again at the end instead of the standalone launcher.
