Next to that basically any burst mage ult will easily hit for 1000+
If we wanna be technical... any execute is the most damage in game as they hit for 9999 to guarantee a kill below the threshold
But..
Generally speaking, mages. Within them, Kukulkan, Zeus and Poseidon come to mind as great damage dealers.
Assassins can dish out a lot of damage really fast, but they are mechanically more complex than mages. Also, they are a bit more build dependent.
But your question is pretty hard to answer - between single target or AoE, ranged, melee, early, late.. easy to handle, one skill or combo... and other parameter.
Zeus, Anoobis, Scylla, Discordia, Hera, Poseidon, Vulcan, Freya... most of Mages can dish a lot of damage in a short time... and i agree for most of Assassin (Pelee, Susano, Thananoob, Bastet...).
Kuku can very easily reach 1k magical power while Ymir will only hit like 200-400
In addition Kuku actually gets to build pen while an optimal Ymir will probably just get Void Stone
Kuku's 3 deals more damage than his ult if the enemy gets stuck in it.
