Also, items that you intend to use often should be equipped to your inventory (that way, you can use them directly during play by switching to that item and pressing R). You probably don't want to fill every slot (this makes it harder to switch back to your estus when you need it), but having a couple extra combat items readied shouldn't hurt for most fights.
also buy a controller they are very cheap..
WASD = Move
Q = Target lock
E = Interaction/pick up

You bought the damn game on pc, so you should be able to use it with pc controls, and not the controller.
To your question, you can use it. Just put it where you have estus, sort through them with one button and then use it the same way you use estus. The game works perfectly with k/m and no need for a damn controller.
