You need to use some of those blood stones.
Blood stones don't count for opening rooms in your Homestead, and they also don't count for entering the Source Temple, and they also don't become inert stones, until you use them.
You need to have turned 12 stones into inert (don't need to carry them around) and then view all homestead memories to get past the door. Any stones after 12 you can keep or save as flashheals during the final boss gauntlent.
the door can also be passed with a physics exploit... you can't throw a teleport pyramid through it from the inventory but if place the pyramid on the ground and then throw it without putting it in the inventory you can toss it through the door with no problem.
