You need two hands but using mouse you could get away with using one finger for LMB. There is aiming, however but it's not very important I don't think and the game, while in combat, will let you quick shot by pressing F when you have your gun out. Quick shots are basically chest shots and generally require two hits on the enemy to take them down unless you have a very good gun and they're a low level.
Combat controls are E, F, SPACE, LMB.
SPACE to break an enemies defence, E to counter attack and F to dodge bullets. Mouse is used for attacking and moving the camera around. All controls are important as the game does require you to use them in most fights(You'll have to break defence with SPACE a lot and E to counter as well as F to dodge bullets). The only button mashing is LMB to attack which you click a lot.
If you were using one hand you could remap things... If you used the keyboard you could probably rearrange the keys so that SPACE is attack, CTRL is free-run up/jump and leave the other controls as is. Sneak mode is now a mode of itself which is very distinct from previous versions. You press C and you're in sneak mode. V brings up Eagle Vision which stays up as long as you don't interact with anything or sprint but you can walk and sneak with it on. No cooldown on it this time around.
