You can find the correct numbers on the wikia.
They don't cancel each other, but I think there is only so much priority value to be distributed between the different inclinations, so when you add to one, the others loose some. If you use enough of one kind of draught, you will set the other inclinations to their minimum priority level. That's probably why you will occasionally see a pawn with only one inclination displayed.
Depending on your inclination status you should drink 2 or more for you main.
Dinput8.dll is a mod that shows the status of your inclinations you could use that to see what they are at.
Just don't use it to change the actual status of the inclinations as it will trigger a ban of your account.

Inclination draughts are LIFO, pretty much, Last In First Out. If somehow your pawn has previous inclinations it may require more to change them to get the order and inclinations you prefer. Supposedly (I have read but not official) there are 4 inclinations, your primary and secondary and 2 more that are hidden. Unless you really want to bother with the last two just ignore them.
If you just want the primary and secondary and your pawn currently has no inclinations (Esc>Status>Tab to Pawns Tab>Profile) you should be able to get away with 1 draught each. Use the one you want for the primary AFTER you use the one for the secondary inclination, then check the pawn profile and verify you got what you wanted. At worse you may have to use another Primary, but I doubt it.
Later he will sell Neutralizing draughts to wipe the inclinations. This is usefull if you already loaded up on inclinations and are really changing your pawn, say from Medicant Utilitarian Healer Mage to full bore raging Scathing Challenger Sorceror.
