No missions to start
I'm new to Mafia III. and I already spent 50 hours playing with it. Then I had no op to pick up any new missions, so I thought it must be a damn bug, but fine, I like the game so I start it from scratch. Now I've got the same issue, after having met with Burke, Cassandra and VIto, got no option even to smuggle dope or booze. Has anyone found the solution yet?

Did you check for missions from the mission tab on the map? It shows all active missions.

Otherwise you may need to zoom in on the map to see mission icons.

AT THE VERY BEGINNING, You have a meeting with Donovan and he tells you about the three potential underbosses. (Cassandra, Vito, and Burke.) Depending on which one you choose to contact first -- that's ALWAYS Cassandra - go to
Escape, map
Lower left corner, Objectives
Choose the Del Rey Hollow panel.

That will pop up a task list you must follow to secure Del Rey Hollow for Cassandra.

I did...Says I can talk to all 3 characters but there is no conversation. I could also steel the semi again, but only the icon appears, the truck is not there. Very odd

The semi-truck mission is bugged to ALWAYS show the map marker. But if you are at max earn for all the districts for that boss, then you cannot do it.

You MUST have that initial conversation with Donovan in his motel room. Once he shows you the three dossiers, you decide who to talk to first. In each case it is NOT as simple as just walking up to them to start a conversation. You have to do some sneaking and peeking to get close enough to start the conversation WITHOUT having to start with a gun battle.

Just look at the main map, Objectives (lower left corner, pick a district (Del Rey Hollow, River Row, or Pointe Verdun), and then follow the directions in the drop down menu. That _should_ get you started.

You all ways start off with Cassandra. After you have taken over Hollow from Dixie mafia, you go to Donovan in hes motel room where he present you with 2 other files (Burge an Vito). After looking one or both of these files, you unlock theyr missions to help them take over River road and Point verdun.

In case of all 3 characters there is no need for sneaking in order to talk to them, each one of them have special objective that must be done.

You don't get missions directly from them, only side missions and thous comes when they have more territory under theyr rule. Everything else (Rackets, capo/lieutenant killings) Comes from either Donovan or some special person that tells you about some rackets business. after he or she have tell you about you get special markings in your map that you need to click. (Like a skull, or exclamation mark). All ways look at your map and zoom closer to see them.

This is pointedly wrong. In Cassandra's case, you MUST _first_ interrogate a Haitian informant at the Taxi Garage to learn where to find the Haitian boss (Cassandra). Then you sneak into her house above the voodoo shop and wait to talk to her.

In the case of Vito, you MUST battle your way past Greco's goons just to rescue Vito. THEN you can have a conversation with him.

Burke is the only relatively peaceful approach. All you need to do is take control of his car and then Drive Like A Maniac to prove that you have the cajones for what needs to be done.

When i usually sneak, i go around the back or something. In this case i went through the front door, breaking it in front of everyone. Not exactly sneaking.

Out of curiosity, during the entire playthrough, how many of your opponents did you whistle to their doom, as opposed to going in guns blazing?

Personally, I VASTLY prefer doing all of my takedowns as quietly as possible. Possibly a 70/30 split, quiet versus violent, overall.

None since i did not know i have whistle. This is my second playthrough since 2016 and even back then i don't remember ever using it.
I did not go gun's blazing however (unless i have to), i was giving to option to use silenced pistol and i use that during the game while doing couple takedowns here and there.

If anything, the whistle (T-key) makes the game way TOO easy. (Along with the Combat View -- V-key -- option.) The combination makes Lincoln like Superman (X-ray vision and superhearing) and the Shadow (nearly invisible in shadows). With those features, even Hardcore (or whatever the mode is called) is practically a cakewalk.

I did not use any of thous. just me and my silencer walking around taking headshots and never getting notice.

It's worth noting that in M3 vanilla, Lincoln didn't have access to a silenced pistol until about mid-game. But when Definitive Edition was introduced, the devs had added the pistol _immediately_. [They had also added a FAST Sports Class car that was actually more of an Exotic.) [Oddly, additional playthroughs didn't necessarily have those two items available. They seem to be randomly made available.]
