When you see a dire wolf model that's scaled up to be bigger, with a glowing cheetos aura around it. That's the alpha.
They haven't implemented it yet, but from the looks of alphas is going to look like something a 4-year old came up with :)

i think is the right click, when im raiding a wolf and i use it i see a counter on the upper right corner for like 50 secs, maybe thats the "alpha" buff (the howl)

Known Information: The species has an incredible affinity for teamwork. Unlike most creatures on the island, Canis is a dedicated pack hunter, and rarely hunts alone. In fact, Canis is naturally as effective with its packmates as most creatures are when their mate is in danger--when in a pack, they are naturally spurred to fight for their lives. Furthermore, when in a pack, the most experienced Canis will be designated "Alpha" and gain an even stronger enhancement.

Taken directly from the dossier. Interpretation would dictate that the highest level Dire Wolf would be the "Alpha" and thus the pack would attack what the highest level Dire Wolf is attacking.

Dire Wolf alphas are NOT like the traditional alphas we have in game currently (raptor, carno and rex). It is merely a asthetic mechanic to coincide with the reality of how Dire wolves and even existing wild wolves behave.

Much like how raptors will always attack a lower level tribe member over a higher level one, or attack a tribe member with a raptor egg when travelling as a group.

Jat mentions that they are not in yet in this thread

I am pretty sure it more of the Pack Leader "Alpha" vs the glowing Alphas we currently have ...

And the Pack parts seems to be working ...

Confirmed by who? Because it wasn't the developers that said that. They said it wasn't in here due to them just wanting to get the patch out, same with the megaloceros charge. Give it time

Basically this would appear to mean that (when it is brought in) sex doesn't matter to get the same bonuses as "mate boosted". Any wolves in the save vicinity boost each other, with the highest level wolf getting a larger than normal bonus.
How / if the howl figures into that, or if the Alpha needs to be a male, is anybody's guess.

There is literally a comment that says they won't be like the original alphas... It will just be a bigger wolf with buffed stats. That's it. There will be other cheeto puff alphas but wolves won't be one of them
