Gameplay should usually take about 15-20 minutes per match. It's longer against certain villains and shorter when clicking through the animations.
As far as game time, it depends on the decks chosen, what cards are played, decision making, etc. But I usually expect to be playing for anywhere from 30 minutes to like an hour and a half or so (again depending on the above factors). There are also options to speed the game up by removing some animations and speeding up the others. Though that might not help you much if you want to play fast in multiplayer.
I find the game only ever slows down if there are lots of targets to choose from (as mentioned above) or if I've been playing for hours on end. <.< That could just be me, though.
That said it only slows down when compiling the list the first time. For example, on any of Infinitor's "deal 1 damage to every target" slows the loading of the list down considerably when he's got 10+ targets in play and Captain Cosmic has roughly the same number. But it only slows down on that initial listing, letting you click and choose which ones to target first and not having to wait for the list to load again. At least for that one effect; a new list is generated for every effect.
As for the ability to fast forward, there's actually an in-game slider for animation speed. I find the maximum speed on card draws and page flips to be a bit excessive, so I just click through most animations instead.
