Based on what I did versus Dethkite and so as to not step on XPW CC's toes, my guess for the unlock is Win a game as Captain Cosmic against Infinitor. Captain Cosmic must destroy at least 5 Manifestations, and must deal the final blow on Infinitor.
If it's not that, it's probably something requiring a certain number of Constructs *and* Manifestations destroyed.
Team was Legacy, TLT, CC, Scholar of the Infinite and Fugue State Parse at Freedom Tower vs Infinitor.
Notable things: I let Cosmic do a lot of damage and break a lot of manifestations and Constructs, as well as score a killing blow. The rest of the team supported as best they could, with Legacy buffing damage while trying to find his ring, Tachyon doing all kinds of AoE and silencing multiple nasty manifestations while giving us all the cards, Scholar was tanking up a storm, and Parse was buffing damage and feeding extra plays to Tachyon and Cosmic. Also found a hilarious appreciation for Ironclad Maintenence Bay, Fugue State Parse and Hypersonic Assault...saved me from some nasty hits multiple times XD
Legacy, TLT, Tempest, Omni-X and CC in MDP.
All constructs were out and subsequently destroyed with Construct Cataclysm; a blow which destroyed Infinitor.
Not much of note other than that to be honest - CC probably destroyed fewer Manifestations than other heroes.
No log I'm afraid, as I was on my phone.
I tried a game with Legacy, F6Tachyon, Hunted Naturalist, Xtreme Prime Wardens Tempest, and Captain Cosmic vs Infinitor in Final Wasteland (given that we've had unlocks in Freedom Tower and Mobile Defense Platform, the environment didn't seem to matter).
Captain Cosmic double skipped all his turns. I got through the entire villain deck, though Machinations of a Madman never entered play. Each type of Manifestation entered play and was destroyed. Infinitor flipped once and back once.
I'm guessing the unlock requires Captain Cosmic to do something or requires Machination of a Madman to enter play. I also noticed that Machination of a Madman was played in every unlock but none of the failed, so my guess is the following:
Defeat Infinitor with Captain Cosmic on the team. Machinations of a Madman must enter play.

Similar set up, but with Ra instead of Hunted Naturalist. Still double skipped all of Captain Cosmic's turns. This time Machinations of a Madman entered play, though Insanity didn't.
I did a bit of analysis on the unlocks. In each of them, all of Infinitor's cards entered play with exception of Whispers of Oblivion, which did not enter play during Tskami's unlock. In failed attempts, including Tskami's most recent, either Insanity or Machinations of a Madman did not enter play.
I'll play a bit with double skip Cosmic tomorrow to try to get all of Infinitor's cards out in one playthrough, just to verify that Cosmic does in fact need to do stuff to get this unlock (or at least rule out Infinitor's plays as being the only requirement beyond the game setup).
