Report a translation problem
Although seriously, if you are looking for the biggest, most important city in Faerun, it would have to be Waterdeep. It's kind of like the Forgotten Realms' New York if instead of a Subway, New York had a giant seemingly endless Dungeon sprawling out beneath it.
Shadowdale is pretty important despite it's small size, if only because Elminster lives there.
Neverwinter has become very popular because of the video games, but I never really heard much about it in the old 2nd edition books I used to game with.
If I recall right, Luskan, the "City of Snails" was the setting of a really good book.
That's about all I remember. I'll post back if I recall some more.
Obviously, a good rogue centric story would work. A massive city is a massive playground for a burglar.
I think the problem with Waterdeep, in terms of adventure, has been how completely vanilla it is. Maybe this criticism is unfair but all the focus on it has only been Undermountain. I think a dungeon master would have a real challenge on his hands if he were to make an interesting story there.

Some clarification:
- Faerun is in the Forgotten Realms campaign setting.
- Sigil is in the Planescape campaign settings.
- Neither setting are the default one; The default setting is Greyhawk (in 3rd Ed - didn't play any other ones)

Anyways, my favorite cities in Faerun are Zhentil Keep and Skullport.
And no, Sigil isn't the best city in D&D, although I do like the Lady of Pain.
The Forgotten Realms are, by far, the best D&D setting there is. Ed Greenwood > the rest.
