play the first mission, quck save your game after you tag an enemy that is about to go behind cover that you can shoot through, kill him with the penetrative shot.
reload the quick save you just did, and repeat. you will get your upgrade fast if that is what you are looking for. (save---- shoot-load-shoot-load...)

Imagine how much time that would've saved me maxing the Luger's recoil if I knew!!
(run like a mad man with extreme heartrate and kill 30 people, (and the mastery skin is killing 10 by shooting their grenades!))
Now I see some quick grinding tomorrow to get the last damn shots in.. had no idea it worked that way
Then there's the really weird thing where you shoot through I-beams and girders, but THAT doesn't register as penetrative. Call me stupid, but THAT sounds like penetration to me!
NOTE: Oddly enough, you get better results when you DON'T compensate for range, when you're shooting long distance. The ballistics physics in this game are REALLY weird, NOTHING at all like real life, and not consistent in-game. But, if you can't get an assisted sight at one position, shift laterally, or move in closer. Snap-zoom-empty lung works best.
