Puncture, as mentioned, from the One-Handed weapon line and Inner Fire, from the Undaunted skill line, are the two skills in question which allow you to function as a tank.
To respond to your "Can I tank with a two handed sword" the answer is really no. In Dungeons and Dragons Online you could, because damage was the main way to tank, but here tanking is pretty much down to using those 2 skills for aggro, then mitigating the damage through blocking, of which a shield will serve you much better as well as the one-handed skill line.
TESO has you using 2 weapons at once, and there are times when both weapon sets will want to be one handed weapons and shields, but others you can switch your second weapon to something else so sure, you could try a two handed weapon. But if you're doing this for aesthetic reasons or because you wanna be a badass tank with a 2 handed the answer remains that this is not the game for doing that.
