So I've just killed the crimson elder. What is the closest door I can go through to get to plant 42?

Just keep progressing through the game. You'll find it.

Plant 42 is in an entirely new area. The helmet key is meant for the third "chapter" of the game so you don't need to worry about it for now.

Just figured the helmet key was the next thing to get. Didn't realise there was another door through the grave yard, painting puzzle room then through the bar door and put the stone and metal thing on the pedastal

youre on the right track. you need to complete a whole another area to get the helmet key. be warned though you may not like it when you come back with it

Why? Because of the hunters?

Mansion > Shack > Guard House > Back to Mansion > Caves > Shack > Back to Mansion > Lab.

That's the flow of the game if you know what you're doing.
