As the guy before me said, it depends on the outside temperature. Personally for an area that size I'd probably set up 6 or 7. Even then, it will probably get above freezing during a heat wave.
Just want to point out that a 20x20 room for food is an absolutely massive area, and your average colony won't need even half that much space. Half the space means half the coolers to keep things cold enough.
Also, one last thing: making all the walls around the freezer double-thick (so 2 blocks instead of 1) helps a great deal in insulating it, and will make things easier on your coolers.

Use the Quantum stockpile mod to make smaller, more effective fridges.
I use a 9 by 5 room with 2 coolers and 3 quantum stockpiles, with a 3 x 5 stockpile.
They do use a lot of power though.
