Cliff Notes answer: there's no specific line in the save file to add an incapability. Incapabilities are added by a pawn's backstory. In order to remove an incapability, you have to change his backstory. (the only exception I'm aware of, is pyromaniacs)
It converts "Can't Do" tasks into "Hated" tasks. So you can technically assign them do to those jobs, but they'll suffer a mod penalty that gets bigger the longer they're doing a hated job. EG, characters with a backstory like Noble, will not want to do menial labor like hauling and cleaning, but if you were in an emergency, say toxic fallout was about to make all your food sitting outdoors waiting for someone to get around to hauling it to rotten, you could have that character assigned to haul for a short while to rescue your colony's food harvest. Just remember to that prissy noble off hauling as soon as the emergency's past.
