Uh... no.
I don't mean "no" because you're running too many mods, just "No" because how in the world can anyone hope to truly diagnose the problem when you're running so many mods?
/sigh
First thing - Where's your log? Go check your log and cut/paste sections of relevant error messages.
And... post your mod list in some .txt file host somewhere like pastebin or something so it doesn't break Steam. :)

Next up - What are you specs? Use Steam to help post them: At the top of the Steam menu, click Help, then click System Information, then copy that and paste the relevant infos, like RAM, CPU, GPU, OS, etc.. That will help get the basics out of the way.
PS - Makes sure you've already updated all your drivers, since that's something many will ask about.
Other considerations that will clean up your game:
Are you using RuntimeGC?
Are you deleting old saves? Deleting save reduce the weight of the files the cloud has to sync in steam so deleting them will improve the sync download when you close rimworld.
I had stutter issues until I removed some mods and ran runtimeGC to clean up the files. I'm also told closing steam after you load rimworld might help as it removes any downoad stutters.
Just wanted to say - Props to you for organizing your OP to provide necessary information and for not immediately screaming at everyone that replies to "fix your game" like so many others might! :)
Check your game logs to see if anything is reported there that looks suspicious. You can also get the game to create a more verbose error log, IIRC, by going into devmode and clicking the report function. (You'll want to disable it after awhile since those logs can get very big.)
What screen mode are you running? (Note: You should be good since you're using an NVidia and not an AMD based chipset, but it helps to check to be sure what mode you're running.)
Your stutter is most-likely mod-related, but you may want to also disable the Steam Overlay if you notice any mouse or display related or resolution related problems. If you're running at high or extended resolutions/display ratios, I'd recommend disabling it. (I disable it by default, but that's just something I always do, since it can cause issues in a lot of ways.)

If so- we may have the same problem and could check between us common mods and suck to find the cause.
...BUT, there's a really good chance it will uninstall several of the hundreds of your mods as it only sorts mods it is aware of. Sending the author your json file afterward will help the tool but it will also cause you some pain with reinstalling a bunch of stuff and manually sorting out where it all goes. If you run RWMS and can then start a game without the problem reoccurring, then at least you have a good starting point...
