Looks like it is now the "AddOptions.txt" file
C:\Users\Name\Documents\My Games\Sid Meier's Civilization VI\AppOptions.txt
Go to options, change anything, click apply, then click RESTORE DEFAULTS on the bottom left. It will say that this will be applied after game restart. When you then relaunch the game, it will recreate all the settings.ini files, and from now on, you are able to save your settings normally.
If you have installed CIV 6 but can't find the file This PC/Documents/MyGames/Sid Meier's Civilization VI/ folder you likely have Controlled Folder Access turned on (or some App or Win 10 update turned it on without your knowledge. This setting prevents CIV 6 from installing the above file folder in order to store your game data and saves.
You can turn it off but at your own risk as there are some security issues involved with allowing programs to automatically write files and file folders to your C: drive.
To turn it off go to Start, PC Settings, Update & Security, Windows Security, Virus & Threat Protection. In the box that opens up scroll down to Virus & Threat Protection Settings, click Manage Settings, scroll down to Controlled Folder Access, click Manage Controlled Folder Access, toggle the Controlled Folder Access switch to Off. Close the menus.
Start a new game and save it. Check Documents/MyGames. File folder Sid Meier's Civilization VI should be there. Open the folder. It should have all the missing data in it, including your your Saves folder and the AppOptions.txt file.
Try \Users\*your*user*name*\AppData\Local\Firaxis Games\Sid Meier's Civilization VI\
