As the game launches default with a 16:9 aspect ratio you will get into trouble if you have a 21:9 for instance. The Button appears to be in the background image and there is a layer ontop of the image that is supposed to be on the same spot.
That remembers me of my first HTML trys 15 bloody years ago. Holy moly and a payed bloody 80Eur for this.
You might be able to hit the button by clicking frantically on the area below the button. If not you have to first change your monitor resolution to 16:9 which will make all your windows windows resize and you have to spend hours fixing that again.
First thing I did was change the resolution and set to windowed, which resolved the problem for me.
It's an absolutely unforgivable bug, tbh. Really obvious, and really ♥♥♥♥♥♥♥ stupid.
Had a minor miracle where it was changing with a flickering screen from alt-tabbing and I was massively mashing and clicking, and it somehow got it to work. Don't know if I can re-create again lol...=/
So looks like I'm never closing the game until they fix whatever it is... And if there's a power-cut (which with my luck, it'll be any minute now), then I'm just going to forever curse Firaxis for this torture.

1. Go to \Documents\My Games\Sid Meier's Civilization VI
2. Edit AppOptions.txt
3. Change this:
As mentioned earlier, there is a game configuration text file here:
1. Go to \Documents\My Games\Sid Meier's Civilization VI
2. Edit AppOptions.txt
3. find the lines below and change 3840 and 2160 to 2560x1440 or 1920x1080 or whatever your monitor can handle.
;Render width in pixels.
RenderWidth 3840
;Render height in pixels.
RenderHeight 2160
Also 10/10 game, spending 30 minutes trying to fix it so it works on boot really adds to the experience
