As for the anthology... I don't actually know what you mean by vanilla civ 6. Like do you have the DLCs (gathering storm and the others)? Those DLCs can change the game a lot by giving you modes that spice things up. Apocalypse mode is a wild ride. I'm pretty sure the dlcs also have community add-ons that implement QoL improvements, better hud, being able more clearly see where your resources are coming from.
I can't give you a proper answer as I haven't played the base vanilla game and then jumped to the complete pack. So I'm actually not sure how much you're missing out on.
An example is uhh, Elenore.
She can be either French or English, depending on the version you want and they are quite differnt from eachother.
Since you are new; these 'small changes' totally change how you want to optimally play any particular faction. Certain ones are very particular in how and where you want to setup while others have more bonus' pertaining to science or culture victories.
Small changes -yes. But these small change have massive impacts on how you will play.

PS: You do not NEED these extra factions but I highly recomend them if you think you will play the game for a long time.
Many people have spent months perfecting certain strategies with particular factions.
Im a big fan of taking faith-based factions and then using the faith to mass-rush units via faith-purchase in the midgame to overwhelm people who dont see it coming by stocking up on faith and then just massively spamming units into armies and then conquoring- From a religious gameplay prior. Can really throw people off. ;) Plus, its fun.
But I digress, these are things you probably dont care overmuch about at the moment and I would definately agree with you on waiting for a sale. They are not needed for you to learn but rather, master differnt gameplay styles.
These are the DLC I would buy if you have interest in the game but BE WARNED; it DRASTICALLY changes gameplay with natural disasters and having a World Congress.
World Congress is another method of winning the game via diplomatic favor.
Gathering storm causes natural disasters like volcanoes to erupt, hurricanes and floods and it also DRASTICALLY changes the game. -These do destroy your buildings and damage your units requiring repairs and heals.

Highest difficulty I've ever played is actually after prince diff so idk....
Usually I go for facism government thus getting dominance victory cuz everytime the game is just going to result into me conquering other civ if they're surpassing me in anyway possible.....well at least my current is focused on culture which im leading ahahaha...
And I've also seen some new features such governors or like loyalty points in cities.
Kinda very new for me but at the same very very interesting for me....I'm very stoked for the sale and the expansion kinda make me feel more immersive.
