Do you have Gathering Storm? GS completely change how strategic resources work. If you don't have GS, you get only a copy of each horse tile you have improved. One copy let you build units that require horses in cities that have an encampment, two copies let you build on any city. Any extra copy above two is for trade and won't give you any benefits. In GS strategic resources are generated per turn and you pay strategic resources to produce, buy or upgrade units that require them. In GS, a improved horses tile will generate more than one (2 iirc) and it will be per turn. Without GS you get one copy and that's it.
It won't, you won't be able to build a district or wonder in a tile that have a strategic or luxury resource. They can't be harvested, ever. You can settle cities on top of strategic and luxuries resources, which will give you the resources as if it was improved. If a strategic resource is revealed under a district or wonder you already built, it will also give you that resource as if it was improved. The resources are never removed, only bonus resources and tile features are removed when you build a district or wonder on top of it. Removed, not harvested. You won't get any yields unless you harvest first with a builder.
Well You Can Build A Plantation On Luxury Resources
The specific comment that was a reply to mentioned Districts and Wonders, not Improvements. Of course you can build the correct improvement on a resource. You cannot build a District or Wonder on one though. Unless a District or Wonder is there and the Resource is revealed later, it's impossible for them to be on the same tile.
