Best way to learn this game?
Jum in with all DLC go?
Or have a vanilla round first?
Also, any usefull youtube video's would be much appreciated.

Jump in with what you have. If you already have DLC great, if not, learn what you've got access to.

For videos, look up the YouTube channel "Aspec"

customize your game

dont take huge galaxy and 30 empires, i recommend small galaxy and 2 or 3 empires

trial and error

save often ^_^

they have a dev clash going on, they are on break now for the holidays but you can watch all the old ones on paradoxs twitch/youtube, and for learning it just time I personaly feel like I was still new at it even after I hit 600h, I am well over 1k now I think lol and they change stuff so you have to relearn the new system when they do so be ready for a new learing curve to pop up from time to time

Trial-and-error? I'll be perfectly honest, I learned this game just from looking around at the interface and seeing what elements there were, how much stuff there was to tinker with, and what building produced and consumed which resources. I never bothered looking up what would happen if your resource count fell below zero, sufficient to say nothing good? It only took me one or two games both pre and post 2.2 to figure out more or less what I needed to do in order to get a big empire going.

vanilla, non iron man, save & reload while testing things. That's clearly the best way to learn.
