Try to install the required software manually (that steam installs when you first start the game). Run these files inside of your steam directory (Usually C:\Program Files (x86)\Steam)
If that doesn't fix it, try to validate the game cache. Right click the game in your library, then go to Properties -> Local Files -> Verify Integrity of Game Cache.

Can you give more details of the issue? Is it crashing with an error or will it just not start? Can you run any other games that use Unreal Engine 3 (For example, Batman: Arkham ###, Borderlands, Mirror's Edge)?
Sorry for the inconvenience, sometimes these engine specific issues are unavoidable sadly :/. Try to reinstall/update the 3 most likely culprits: DirectX, VC++ Redist and .NET Framework 4 and report back with more details.

Go to:
Steam\SteamApps\common\A Story About My Uncle\Binaries\Win32
and find "ASAMU-Win32-Shipping.exe" application right click to "Properties" and "Compatibility"
Check Compatibility Mode and Run as Admin.

right clicking to 'properties' and going to 'compatibility' (as DƆ.rickajoie said)
running the compatablility troubleshooter
clicking 'troubleshoot program'
checking 'the program worked in earlier versions of windows but won't install or run now'
clicking 'windows 7'
clicking 'test the program'
it should then work, and then you go through the rest of the troubleshooter, clicking 'yes' when it asks you if you want to save the settings.

ASAMU-Win32_shipping.exe has stopped working
a problem has caused the programme to stop working correctly. windows will close the programme and notify you is a solution is avalible.
side note- i am using the intel hd 530 in built graphics as i dont have a gpu in my custom build yet and i have had a problem running battlefield 3 and scrap mechanic due to the fact i do not have a dedicated gpu. is this possibly the case???
