if you are on steam beta - opt out, Known to cause issues in past.
Possibly onwards to sfc /scannow to be sure.
Else, approach from the other direction - and figure out 'why' storymode mods aren't working. Assuming 'they' were installed correctly.... (and that vanilla/clean gta5 works fine)
I'd be leaning towards corrupt 3rd party requirements like vcredists, .dotnetframework, av/security (you say av is disabled --- but that doesn't 'always' mean it is ALL disabled' - especially if a 3rd party - now disabled and windows defender steps in - hell something keep messing with things til they are outright uninstalled (and some even need special uninstallers/cleaners for that tooo)). This could also include controlled folder access if enabled, other exploit protection settings if not defaults, and whitelists you could use. Disabling overlays... trying with r* launcher, game, or even steam as admin...
...and producing the log files / crash reports / error codes...
