Browse to the root of your GTA V installation folder and open the file "commandline.txt". If it doesn't exist then create it. Then add in "-DX11" without quotes in a new line in the text file and try running the game now.
2. Make sure that your settings.xml isn't "read only"!
3. If 1. and 2. didn't work, try to verify your gamedata and try both steps again, this helped me to get rid of vsync, which I couldn't get deactivated over the ingame settings.
