But to answer your question, only thing you need to scope out is the primary target. Other than that, you can scope as much or as little as you like. You can always come back and rescope it out before the heist if you choose.
I usually just only scope out until I find the secondary targets I want then I'll leave. Two stacks of coke, 4 stacks of gold, etc.
On the other hand, it is not a bad idea to scope out the full island and unlock everything possible. Unless I am missing something, with the exception of the loot and antenna everything else stays scoped out forever... so you might as well do it on your first or one of the early runs.
