Do I have to host the mission for the npc to call me for new missions?

you can play all of them simply through the use the online jobs function.
esc>online>jobs>ply jobs>rockstar created>missions> casino missions. (for example casino- loose chang)

They work simply as contact missions. you need to be ceo/mc/vip if you want to start them from the casino. It was a little bit rough to become host at the first day but i think they fixed some things and it works better now.
There is an achievement for both, playing every mission as crewmember and host, each grants 100k.

all in order achievement for 500k.
play them in the correct order.
loose chang>housekeeping>strong arms tactics>play to win>bad beat>cashing out.

you can find all awards under online>stats>awards>casino dlc to check your progress.

you get a free car "enus paragon armored" for finishing the host and crewmember achievement.

I don't see any casino mission on the mission list at all.

if you want chips beside the 50k daily limit do agatha freeroam mission each one grants 5k chips. Simply call her while being ceo/vip/mc and request work

it was pretty awful to attemp becoming host right after dlc release, the servers were melding under the high load.

for me it was like this
i bought the penthouse > entered for the first time the casino> got the tour from the manager > met agatha for the first time in the penthouse>left the casino >got a call from her to revisit her at the casino for special work> entered the casino got a special sign of her, which is only available while being ceo/mc/vip to enter and was able to choose all casino missions in the job list.

i dought you need to own a penthouse to host the mission but iam not sure about it.

for me it was like this

Well i don't own a penthouse so this is the only difference between us

That's your issue. You need a penthouse for "silver status" so you can host missions.

you don't need the house to get the 500k, only someone with the option to start the missions in the correct order or simply join mission in the correct order of randoms
reward for hosting all missions is only 100k
