Are there supposed to be quest that lead you through the game?

I think I did maybe 6 quests and forgot to turn in one of them (Shock Therapy?) and just kept playing. Killed the first boss, noticed I had a quest to turn in, ran around until I found where to turn it in and after that I haven't seen a single quest anywhere.
I'm fine with there not being quests, just want to make sure I'm not missing anything important.

I remember my first playthrough got bricked because I did not turn something in before completing another quest, pre 2.0 though
