Can secret rooms pop up in the quest to kill the Garden Guardian?

I took a hit to my hero roster today and find myself in need of another up to level hero. Problem is, only source is the secret rooms in the courtyard that let you rescue a level 5. Can those rooms show up on the Garden Guardian's boss kill mission?

So far as I know, no. I've never seen it, and I've done lots and lots of Garden Guardian missions.

If you have CoM there is also another rare event that lets you buy an L6 hero for 20 shards.

Bringing a level 0-1 character to a short champion mission with the ancestor portrait is always an option, if the team is well made and you can prevent surprise attacks the task is less suicide than what may seem.

Just make a Man at arms babysit the new one with guard+ riposte and have like
Hound-Occ- XXX - MaA as an easy pull-through team comp.
