If you are new to the game, look at the guides section. There are 5-6 really good general ones.
In order to have a good chance to succeed on a mission you need:
1) A team that works well together.
2) The appropriate amount of supply items.
Read the guides, or just figure it out yourself by trial an error.
If you want an easier game, try Radiant difficulty, and you can also go into settings and make it even easier.
-Bring a party that is good with shuffles, you'll see why on the boss
-Definitely bring a hero for stress management. I always opt for a crusader
-Bleed resistances. Nuff said.
Well, this party has no stuns and no reliable healing, so I'm not surprised you ran into some trouble.
A few things that help...
-Bring max bandages. You don't need antivenom, but you definitely won't regret bringing these. Having four holy waters for the boss is nice, but not essential.
-Bring a plague doctor with Blasphemous Vial + Speed trinket (ancestor musket or similar). You need to be able to stun spam the witches and priests before they get a turn. Being able to treat the big bleeds before they proc is also very helpful even if you have bandages.
-Stress healer is helpful in this area since there are two fights where you can stall indefinitely to completely restore all of your health/stress.
-Bring a team that can deal with repeated shuffles. Riposte is helpful for the boss.
-Enemies have enough HP that you need 2-3 hits from big hitters to be able to clear them out. Make sure that your party is synced that all of your primary attackers can hit the same position.
I would go with something like PD/Occ/HM/BH. Front three have big damage mark synergy and good stun options. PD is for crowd control. HM can move back and stress heal when needed. For the boss, bring skills that can target from alternate positions... put Incision on your PD, stab on your Occ, Caltrops on your BH and you'll be fine.
Also, you'd normally want to kill the priests as soon as possible. Their "the finger" attack can hurt your back rank people very badly.

Wait, they don't call for reinforcements unlike in other dungeons?
Don't bring shovels or keys for any of the Darkest Dungeon levels; you won't need them (save your money and inventory space!) For DD1, be sure to have a party that is viable even when shuffled out of position (so no Lepers or Arbalests here). For the first level, as others have pointed out, a Jester can help immensely as stress damage will be a major issue throughout.
Also consider that if you are playing with the "Never Again" option (set by default), you won't be able to send the same heroes into the Darkest Dungeon more than once... and even with this option disabled, as it is on Radiant, it still isn't a good idea to reuse the same characters in different levels. Thus, be sure to have multiple level 6 heroes of your preferred classes for each mission.
In terms of supplies, bring maximum amounts of food, bandages, holy water, and torches. The maps themselves are fixed and revealed through exploration, like those in the Courtyard on in Brigand Vvulf's dungeon. If I recall correctly, nothing nflicts blight or horror in DD1, so no need for Laudanum or Antivenom.
