Is it possible to heal or remove negative quirks?
My crusader steals everything with his cleptomaniac quirk :(

Yes. That's one of three things the Sanitarium does. The other two are removing diseases and making positive quirks permanent. All three cost money and it takes a week so make sure you're curing someone you don't need for the next quest. You should also only cure really bad quirks since you need your gold for more important things.

Alternatively you can use a torch on the scroll pile curio in the Warrens, medical herbs on the corals in the Cove or holy water on eldritch altars that can be found in any region to cure a random negative quirk.

Not unlocked sanatorium yet. How do I tell what quirks I can reinforce or remove by what building or section of a building? Or is it just the sanaterium and the other buildings just for stress relief?

Not sure what you mean. The Sanitarium is the only building where you can remove and reinforce quirks. You can only lock in up to three positive quirks per hero and remove negative quirks as much as you want as long as you can afford it.
