I played RE4 most of the time with the keyboard and mouse. RE4 allows players to rebind the keyboard keys and toggle negative acceleration off from the mouse (modern aim mode).
That's why I recommend keyboard and mouse. I tried controller but I thought that it was awful.
Personal preference is the only variable here.
However, I personally use only mouse and keyboard, and find it to be a more reliable method of aiming and moving.
Pretty much, if you're generally one to play with a controller on games that allow it, play with controller. But if you generally opt for mouse and keyboard, that's probably the better bet. Just like Dekonega said, it all comes down to what you prefer. Game's good with mouse and keyboard, and from what I've heard it plays well with a controller, so just try them both and see what fits your fancy :)

All the needed controls are conveniently placed for the ease of reach. Plus, it also has rumble so, even a more immersive experience. The feeling of Input LAG while aiming, if any, (and usually there is... ) it's also greatly "reduced" as a side effect of the nature of Analog Stick's movement being translated into a speed vector and smoothed out, rather than a more direct 1:1 "per dot" translation as it would be the case for a Mouse driven Aim reticule. It's a more "over the hand" method, but with a bit of practice it becomes natural enough.
First PC port i used controler as well, mapping it at most simillar to GC as possible.
With this port i use mouse and keybord and its really good, except for QTE's, so i would say controller for You if you own one. But if You just plannig to buy it for this particullar game, well its not necessary.
If i made any mistakes, im sorry, i don't have opportunity to use english to often.
Having played the game prior on GameCube and PS2, I have to say the controller works better for QTEs. But aside from that gripe (and honestly, Resi 5 is far worse for QTEs I think), I think the control is very nice with kb/m.
