Kill Esau and quit out to the menu when his death animation starts. Load back in via continue and Esau will be gone for the rest of the floor.
He's the hardest character in the game but there's still a good amount of outs for him.
-Dark Esau doesn't damage other players.
-Even if you die, ghost gonna chase you, meaning as a ghost baby, you can kite him around your enemies and beat the devil out of them.
-When you finished the floor and Dark Esau still alive - kill him. With that, you able to make unreachable pickups - reachable, but make sure you full clear the floor.
-Not sure if you know, but you can release Dark Esau earlier if you press again Anima Sola. More strats with that.

Then I would consider the Repentance co-op as a legitimate cheat, judging by that words.
But that's a good way not also have fun, but breeze through the hardest completion marks.
Honestly with Jacob you need to just stay calm when playing him and don't get yourself killed. Easier said than done of course but a lot of people panic with Dark Esau around and take unnecessary damage.
After Esau dies (which he will), then make full use of Anima Sola whenever you can. Seriously, this item is nuts when you're not restricted to using it on just Esau. 5 seconds of stunning a boss and getting uninterrupted damage on them is pretty good. If you want to play it safe you can always use Anima Sola and then focus solely on dodging while it recharges. Adding to this, 9 Volt is also really good with Jacob; it turns Anima Sola into 5 seconds of stun on a 7.5 second recharge timer which is obviously amazing.
The guy has 250 health, so if you can count your damages on him, and then use Eraser, he stops existing.
Of yourse, if you really need the unlocks now, just hold "R" until you get a top-tier defense item (Holy Mantle, Gnawed Leaf, Meat Cleaver - which is probably a bug, etc). Also whatever you do, don't walk into the Hush door or Mega Stan button with Esau on your back - also an oversight, i assume. Half the tainted characters feel unfinished or barely tested.
1 - Kill Esau, quit the game before his animation finishes. Load back in and he won't bother you.
2 - Find eraser, damage him enough then finish him off with it.
3 - Use Meat Cleaver to create 2 Mini Esaus and kill both. Mini Esaus don't turn you into the lost with the explosions.
4 - Kill him, become the lost and play an alternate version of the lost with Anima Sola.
