You can let queens be made -- that's not a problem. It's likely that you did miss a beat (perhaps at the start of the fight) and didn't notice.

Does that include not missing a beat even prior to opening the door? Because yes I did take a few seconds to pump myself up for the chess fight before opening the door as I had already lost my modifier prior to entering the level.

I think Conga's the easiest boss, personally - if you get into a good spot between the lines you can just alternate attack directions on every beat to clear the conga lines, and then he himself is so straightforward about charging you down. Death Metal and Deep Blues start off fairly easy, but seem to grow notably more dangerous based on how late they spawn (more dangerous adds), unlike Conga who is basically the same fight regardless.
Edit: Nevermind, I'm wrong. I took bard to seeded hardcore and fought the first boss flawlessly both times, except before beginning the second fight I purposely tapped a wall to lose the combo before opening the door.
