EDIT: Second thought, what do you mean by co-op? If you mean both on the same team, no.
Both players can pick a team size between 1 and 3 and switch between characters during a 1 vs 1 match or send in AI-controlled team members to do an assist attack and then leave again, if they have a team of 2 or 3 characters. Or both, just not at the same time (the red/green light indicates whenever you can switch characters/call an assist).
