Yes, beating Marie 300% will unlock all artwork. I've done it before.
The way I beat her was this: for the first phase, get her in the corner and use j.MK x 1 > j.HK > jump > repeat. For her second phase, get her health lower than yours, and keep jumping/double jumping behind her, crossing her up so she cannot hit you, until you time out. Since you have higher health, you'll be awarded the win.

Back in July-August 2013 when the PC version was still beta-only + available only for backers, story modes were enabled in said beta. Squigly's story wasn't finished though + even when it was, the developers wanted to make it available for backers and non-backers alike on PC's official release day.
So, they could have just left the slot for Squigly in story mode empty, but instead they decided to make a joke story consisting of some non-serious dialogue, a fight vs a significantly buffed Marie and a reward for beating her in the form of some Squigly story teasers.
This story mode was later replaced with the normal one, but since some fans enjoyed the challenge of the "Squigly vs Marie 300%" fight, it was kept in extras for everyone who wants to give it a go.
