I've driven a Class 90 HUD but not an ADV. But followed instructions to the letter.
The Loco is an AP_Waggonz Class 90 Pack Virgin U/B.
The Route is WCMLN and the Scenario is "Hand Over", a Workshop Scenario.
I forgot to mention, whilst the train is sitting therethere's a high pitched rapid bleeping, which I can't stop.
you forgot to mention the most vital part of your problem lol
AWS, hit Q to stop the bleeping noise (I think, if q doesn't work try numpad enter)
Which one of these screenshots best represent your cab on starting?

And I've tried Q but not tried Numpad Enter.
I've eventually got the train to move. I think it was because I drained the brakes fully. I also held the Parking Brake off button down for 15 secs after turning it off (RTFM?), except it doesn't say that in the instructions, just the label on the PB.
The problem is, though, that even though the Train Control is set at 105 mph I can't get it to accelerate to more than 38 mph, and that too miles.
And I can't get the Dynamic Brake to less than 57%.
Still bleeping, though.
Do you have any red lights on the LED status lights?
There is an ETS on key to the right of the clipboard
Besides that, it's hard to say from that screenshot.
It seems like you do have a brake on, question is which one. Loco, train, parking. Try turning all the brakes you can think of and find to off. And see if that help.
I'll download the scenario and have a look at it myself.
