For reskins you need to have the base pack, the product info will tell you that. everything else works out of the box. It is a good source for quality extra rolling stock made by 3rd party developers.
There are also some scenario packs. If you're interested in one of these, carefully read the requirements on the store page, as they always need other DLC to work correctly.

Basically it always depends on what you like. You said, you would be new to TS, so I would recommend you play the game for several hours and then decide for yourself, what you like the most:
- American trains
- british trains
- german/austrian/swiss trains
- japanese trains
- chinese trains
- others
Also an important factor is what you like most: steam trains, diesel trains or electric trains?
Do you like more commuter, freight, high speed services or shunting? Modern day or past times?
You see, there's a lot of variation, so it really depends on what you like most.
