Western Mainlines by itself is very long.
I'm not sure if any US or Canadian routes are as long, but they are commonly slower because of speed restrictions.

Canadian Mountain Passes is 159 miles, takes almost 6 hours.
The nice thing about these is a good mix of quality and quantity. CMP is slightly more versatile in terms of surroundings, but Donner Pass has quite the transition as well, took me a westbound full run during Le Mans 2018 to realize.
The Pacific Surfliner is somewhere in this area, too, 130-150 miles, 3 hours for a stopping service. Freight speed limits are lower (4+ hours?) although it doesn't really happen.
There was a recent South African freight route which spans over 200 miles. Not really 2020 quality expectations but sounds interesting anyway.
Western Mainlines and ECML were mentioned with 300 and 393 miles respectively.
^ As Darloman says , The Western Mainlines ( Just Trains ) with the various add- ons , is the longest route I have / am aware of . Penzance to London = 6 HRs game time / driving .
